Is it possible to install and run windows applications through wine without a ntfs or vfat partition?

Yes, that is one of the main reasons for using wine.
The speed is fine, not noticeably slower than a native program (in general that is, YMMV). Make sure you don't fall into the trap of thinking that Wine is an emulator.
I highly recommend you use the winesetuptk program (emerge winesetuptk) and Frank's Winetools to get Wine set up.
