I'd like to change my ServerLayout to go from Twinview to Single and back sometimes, but I can't find a way to do this without chaging the DefaultServerLayout in XF86Config-4.

From the XF86Config man page I found:

A ServerLayout section is considered "active" if it is referenced by the -layout command line option


I'm guessing this is executed something like X -layout, but I really don't know where this goes, or how to go about doing this the 'right' way.
